Unless you're legally blind, then yes you will notice a huge reduction in sharpness going from the 5k 27" UltraFine to a 49" 1440p. The 5k display is 5120x2880, so it could also be called a 2880p display. The 5k display is 218 PPI (pixels per inch), while a typical 49" ultrawide is 108 PPI. That's a quarter of the pixel density.

Another way of thinking about it, the ultrawide is similar to two 27" 2.5k displays side by side.
